For me it must be The Sons of Katie Elder This is because my late Mother took me to see it while on holiday in the late 60's anybody knows the count of shots he gets out of his 6 shooter without reloading in the final shoot out in this film, got to be in the 30's

My view:
His best performance ever, and the best movie overall in which he appeared: The Searchers.
My favorites: The Duke and Maureen O'Hara -- Rio Grande, The Quiet Man, The Wings of Eagles, McLintock! and Big Jake.
Now The Quiet Man was actually a great film, whereas McLintock! was pretty silly. Rio Grande, Wings and Jake were good (Rio Grande the best of those three.)
But those two together -- magic.

The Searchers is my favorite, I thought he was at his best when he played darker characters.

Rio Bravo is a close second. I always used the Rio Bravo test for new girl friends; if they didn't like it our days were numbered. I believe Quenton Tarantino invented that test.

The Man Who Shot Liberty Valance is way up there and I believe The Shootist was probably his best performance; should have recieved an Oscar in my opinion.

I don't mean to hi-jack the thread but I love westerns and some of my favorites are:

The Man From Laramie

Naked Spur

Ride the HIgh Country

Seven Men From Now

High Noon

Outlaw Josey Wales
