I have read that most aromatic blenders use prime Virginia and Burley tobacco. True or false? Does it matter?

IMHO, I think it has to matter because ultimately it is the tobacco that provides the smoothness and taste of a smoke.

I would think so, with the leaf being treated in various ways (a la canvendish) or cased and/or topped. You do occasionally see aromatics with Latakia, Perique, dark-fired Kentucky and/or Oriental Leaf, but it seems to be the exception to the rule. The amount of "tobacco" flavor can really vary with aromatics in my experience.

Of course they do. Some like to look down their noses at companies like Sutliff and say they use inferior tobacco without realising they manufacture many of the Hearth and Home blends among others.

Add Peretti to that list. They make the Watch City flakes, too. And there are other companies who use their tobaccos in their blends as well.
